Microcapsules of Poly(3-Hydroxybutyrate) for Prolonged Protein Release
The aim of the investigation was to develop a system for prolonged protein release based on the use of poly(3-hydroxybutyrate) (PHB) microcapsules loaded with bovine serum albumin (BSA).
Materials and Methods. To develop microcapsule we used microbiologically obtained PHB by a strain-producer Azotobacter chroococcum 7B. Microcapsules loaded with model protein: BSA — were produced by two-step emulsification “water/oil/water” (W/O/W). We applied spectrophotometry, confocal and scanning electron microscopy to examine the morphology of the microcapsules, and BSA loading and release. In vivo biocompatibility of the microcapsules was studied in accordance with intramuscular implantation and histology findings.
Results. The study of BSA incorporation and sustained release from microcapsules for more than 190 h demonstrated the efficacy of the proposed system. The mechanism of protein release from the microcapsules was found to occur due to the rupture of polymer walls. Moderate tissue response to the implantation of the microcapsules obtained was demonstrated.
Conclusion. The developed PHB microcapsules loaded with BSA are a good example of production of long acting protein drugs.
